Description

FOXO4-DRI is a D-amino acid retro-inverso peptide that mimics the helix-2 domain of FOXO4 transcription factor and competitively interferes with the physical interaction between FOXO4 and p53 tumor suppressor protein — an interaction that is selectively upregulated in senescent cells to sequester p53 within the nucleus and suppress pro-apoptotic signaling. By disrupting this complex, FOXO4-DRI is hypothesized to restore p53-mediated apoptosis specifically within senescent cells while sparing healthy proliferating populations.

In vitro studies in human fibroblast and cancer cell senescence models and in vivo rodent aging experiments have examined its efficacy in reducing senescent cell burden and attenuating senescence-associated secretory phenotype (SASP) cytokine production.
